Cloning and sequencing analysis of the codling mothCydia pomonella COⅠ gene

Abstract:

    The mitochondrial COⅠ gene fragment of codling moth Cydia pomonella ( L. ) specimens from the Alar area ofXinjiang was amplified via PCR and the PCR products were purifiedclonedandsequenced. 709 bp nucleotide sequencesere obtained with a very high A + T content (68. 7% ) and relatively low G + C ontent (31. 3% ) . Comparing the COⅠsequence of the codling moth with that of several other moth species,the homologies of nucleotides ranged from 85. 4% to88. 1% and the genetic distances ranged from 0. 130 to 0. 162. Reconstructing the molecular phylogenetic tree of theTortricidae with the Neighbor-Joining ( NJ ) method produced a result in accord with the raditional morphologicalclassification. The results of this study provide an importantbasisfortherapididentification of C. pomonella with DNAbarcode technology.
