Analysis of PCR-RFLP data on the COⅠ-COⅡ gene of three barkbeetle species (Coleoptera: Scolytidae) by microfluidics

Abstract： PCR-RFLP was used to amplify and analyze the COⅠ-COⅡ genes in the mtDNA ofDendroctonusvalens LeConte，Hylurgops longipilis Reiher and Ips acuminatus Gyllenhal． Specific restriction enzymes were screened bycomparison of restriction recognition sites． The amplified gene fragments could be analyzed quickly by microfluidicsfollowing enzyme digestion． The results indicate that this technique can accurately and rapidly identify these three beetle species．
