香加皮乙醇提取物对草地贪夜蛾生殖行为的影响

【目的】 利用植物次生代谢物调控害虫行为是害虫绿色防控的重要措施。通过评估香加皮Periplocae cortex乙醇提取物对草地贪夜蛾Spodoptera frugiperda生殖行为的影响,为下一步筛选干扰交配的活性成分和开发新型害虫嗅觉行为调控剂奠定基础。【方法】 室内采用熏蒸法测定了香加皮乙醇提取物在0.09-14.40 mg/mL浓度下对草地贪夜蛾雌成虫求偶交配的时间节律、次数、持续时间、精包数量和单雌产卵量的影响,利用罩笼选择法分析了喷施香加皮乙醇提取物对草地贪夜蛾雌成虫寄主产卵选择的作用。【结果】 香加皮乙醇提取物暴露处理对草地贪夜蛾雌成虫求偶行为无显著影响(P>0.05),但可抑制其交配行为,其中,7.2 mg/mL浓度处理的交配次数、交配持续时间和精包数量分别为(1.92±0.35)次、(152.50±23.42)min和(1.44±0.13)个,显著低于对照组雌成虫的(3.67±0.45)次、(315.00±70.60)min和(2.27±0.11)个(P<0.05)。香加皮乙醇提取物处理,可显著降低草地贪夜蛾雌成虫的平均单雌产卵量(P<0.05),其中7.2和14.4 mg/mL浓度处理的平均单雌产卵量分别为(586.66±50.32)和(503.63±66.83)粒,分别是对照组的58.19%和49.95%。罩笼试验结果显示,喷施香加皮乙醇提取物对草地贪夜蛾雌成虫产卵选择表现出高浓度驱避和低浓度引诱的生物活性,其中14.4 mg/mL浓度处理雌成虫在玉米植株上的平均单雌落卵量占同一网笼玉米植株上总卵量的31.92%±9.09%,而0.09 mg/mL浓度处理的玉米植株的落卵量则占对应网笼卵量的64.95%±4.47%。【结论】 香加皮乙醇提取物可显著抑制草地贪夜蛾的交配行为,减少有效交配次数,并降低雌虫的产卵能力,对草地贪夜蛾雌成虫产卵寄主选择具有明显的干扰作用。

英文摘要:

[Aim]  Utilizing plant secondary metabolites to regulate insect behavior is a crucial pest control strategy. Investigating the effects of ethanol extracts from Periplocae cortex on the reproductive behavior of Spodoptera frugiperda can offer a foundation for subsequent screening of active ingredients that interfere with mating and the development of innovative pest management based on olfactory behavior regulators. [Methods]  Indoor fumigation was used to determine the effects of different concentrations (0.09-14.40 mg/mL) of P. cortex ethanol extract on the timing, frequency, duration of S. frugiperda courtship, as well as the number of spermatophores in copulatory pouch and eggs laid. Additionally, we investigated the effect of spraying P. cortex ethanol extract on the oviposition selectivity of S. frugiperda in an indoor cage. [Results]  There was no significant effect of the different concentrations of P. cortex ethanol extracts exposured on S. frugiperda courtship behavior (P>0.05), while their mating behavior could be inhibited. Among them, the mating frequency, duration, and number of sperm packets treated with a concentration of 7.2 mg/mL were (1.92±0.35) times, (152.50±23.42) minutes, and (1.44±0.13), respectively, which were significantly lower than those of female adults in the control group (3.67±0.45) times, (315.00±70.60) min, and (2.27±0.11) times (P<0.05) The ethanol extracts of P. cortex significantly reduced the number of eggs laid of S. frugiperda (P<0.05). The average single female egg production of the 7.2 and 14.4 mg/mL concentration treatments were (586.66±50.32) and (503.63±66.83) eggs, respectively, which were 58.19% and 49.95% of the control group. The results of the cage test showed that spray application of the P. cortex ethanol extracts significantly influenced S. frugiperda oviposition selection behavior, with an attracting effect at low concentrations, but a repelling effect at high concentrations. The number of eggs laid on maize plants treated with 14.4 mg/mL accounted for 31.92%±9.09% of the total number of eggs counted in the treatment cage. while the egg laying amount of maize plants treated with 0.09 mg/mL concentration accounted for 64.95%±4.47% of the corresponding cage egg laying amount. [Conclusion]  Ethanol extracts from P. cortex can significantly inhibit the mating behavior of S. frugiperda, reducing both the frequency of effective mating and the oviposition ability of female S. frugiperda. Simultaneously, these extracts have a significant repellent effect, interfering with the oviposition selection of female S. frugiperda.
