细菌介导RNAi研究甘蔗二点螟蜕皮调节转录因子CiHR3基因功能

中文摘要:甘蔗Saccharum officinarum L.是热带和亚热带地区种植的主要作物之一,也是许多国家和地区重要的糖料来源。为了解决甘蔗在农业生产中常常被甘蔗螟虫的为害,重点控制甘蔗二点螟Chilo infuscatellus是一条显著提高甘蔗产量和品质的有效途径。近年来,RNA干扰技术已发展成为控制虫害、提高作物品质的新方法,通过dsRNA诱导的RNA干扰可以用来研究昆虫功能基因。本文以甘蔗二点螟蜕皮调节转录因子CiHR3基因为靶标,建立细菌介导的RNAi方法表达CiHR3基因dsRNA,通过提取靶标基因dsRNA,喂饲甘蔗二点螟3龄和5龄幼虫,结果显示:3龄甘蔗二点螟幼虫在一周内发生形态显著变化、体重明显下降;5龄甘蔗二点螟幼虫在一周内形态也出现显著变化,更明显的是出现畸形蛹,非正常化蛹。本研究结果验证了昆虫蜕皮调节转录因子CiHR3可作为甘蔗二点螟生物防治的靶标基因,为后续植物介导的RNAi表达dsRNA防治甘蔗螟虫打下很好的基础。
英文摘要:Sugarcane, Saccharum officinarum L., is one of the major agricultural crops cultivated in tropical and subtropical regions and as well as an important source of sugar in many countries and regions. In the agricultural production process, one of the factors which are responsible for the low yield of sugarcane is sugarcane borers. Sugarcane stem borer, Chilo infuscatellus, is one of the most injurious pests of sugarcane. In recent years, RNA interference technology has become a new method for protection against insect pests and improvement of crop quality. RNA interference induced by dsRNA can be used to study function of insect genes. In this research, the molt-regulating transcription factor CiHR3 was selected as the target gene. And sugarcane stem borer was fed CiHR3 dsRNA expressed in bacteria. The results showed the different instars of the sugarcane stem borer larvae has become significant abnormal and weight loss in one week. This study is aimed at researching the role of CiHR3 gene as the target gene of biological control, which causes sugarcane stem borers could not molt or their growth and develop process become abnormal, and is used as a biological control tool of sugarcane stem borers.
