【目的】 为了弄清南美番茄潜叶蛾Tuta
absoluta (Meyrick) 幼虫肠道细菌的组成结构及多样性,为其综合防治研究提供理论依据。【方法】 采用单分子实时测序技术(SMRT Cell)对南美番茄潜叶蛾3龄幼虫肠道细菌16S rDNA全长进行测序分析。【结果】 从南美番茄潜叶蛾3龄幼虫肠道样本中共测得25 903条Effective- CCS,聚类为134个OTU,共注释到14门22纲53目75科103属118种的细菌。其中,变形菌门(Proteobacteria)为优势菌门,占93.95%,欧文氏菌属(Erwinia)为优势菌属,占91.82%,Erwinia
iniecta为优势种,占91.82%,其余种类的相对丰度均低于1%。南美番茄潜叶蛾3龄幼虫肠道细菌多样性指数ACE、Chao1、Simpson和Shannon指数分别为77.308±13.489、77.821±12.771、0.156±0.039和0.835±0.190。【结论】 南美番茄潜叶蛾3龄幼虫肠道细菌的群落物种组成丰富,其中Erwinia
iniecta为优势种。该研究结果可为南美番茄潜叶蛾幼虫肠道细菌功能的挖掘及该害虫综合防治研究提供理论依据。

[Objectives] To investigate the community structure and
diversity of the gut bacteria of Tuta absoluta larvae. [Methods] Single-molecule real-time sequencing
techniques were used to sequence all regions of the bacterial 16S rDNA gene in
the gut of third instar T.
absoluta larvae. [Results] A
total of 25 903 Effective-CCS and 134 OTUs were obtained and annotated into 14
phyla, 22 classes, 53 orders, 75 families, 103 genera and 118 species. The main
phyla, genera and species were Proteobacteria (93.95%), Erwinia (91.82%)
and Erwinia iniecta (91.82%); other species comprised less than 1% of the total. Ace, Chao1,
Simpson and Shannon indices were 77.308±13.489, 77.821±12.771, 0.156±0.039 and
0.835±0.190, respectively. [Conclusion] The gut bacterial community of third
instar T. absoluta larvae
has high species diversity and the dominant species is E.
iniecta. These results could
provide a theoretical basis for investigating gut bacterial function and for
the comprehensive control of T. absoluta larvae.
