中文摘要: 本研究运用PCR-RFLP 对红脂大小蠹Dendroctonus valens LeConte、长毛干小蠹Hylurgops longipilis Reiher和六齿小蠹Ips acuminatus Gyllenhal mtDNA 的COⅠ-COⅡ基因进行扩增。通过酶切位点的分析比较,筛选特异性的内切酶,结合微流控芯片技术对酶切产物进行快速检测。建立3 种小蠹的准确、快速区分鉴定方法。
英文摘要: PCR-RFLP was used to amplify and analyze the COⅠ-COⅡ genes in the mtDNA ofDendroctonusvalens LeConte,Hylurgops longipilis Reiher and Ips acuminatus Gyllenhal. Specific restriction enzymes were screened bycomparison of restriction recognition sites. The amplified gene fragments could be analyzed quickly by microfluidicsfollowing enzyme digestion. The results indicate that this technique can accurately and rapidly identify these three beetle species.
