Rapid discrimination of four common species of store booklice, Liposcelis, from China using PCR-RFLP

Abstract:Rapid discrimination of four common species of store booklice, Liposcelis entomophila ( Enderlein) , L. bostrychop hila( Badonnel) , L. decolor ( Pearman) and L . p aeta Pearman was conducted by using PCR-RFLP. Genomic DNA was extracted from booklice with CTAB. Genes of 16S rRNA were amplified with specific primers and PCR products were digested with 5 restriction endonucleases respectively. The results showed that the PCR product was estimated to be about 500 bp in length, which could be digested by DraI . All the booklices examined could be discriminated according to the relevant banding patterns. The results were not affected by geographical populations, developmental stages and sexes of the four species. It is suggested that this method is reliable for rapid discrimination of the four booklice species.
