The comparison of colorimetry and microtitration methods for defermining the protein content and activity of phenoloxidase in insect hemolymph test

Abstract:Protein content and activity of phenoloxidase(PO) were assayed using microtitration and colorimetry, and the inhibitory effects of salicylidene-4-nitroaniline on PO activity were also compared by these two methods. The results showed that there were no remarkable differences between these two methods in measurement of PO protein content and activity, and effects of inhibitor (salicylidene-4-nitroaniline) on PO activity. We conclude that microtitration assay is as good as colorimetry, and is even better because it is simple and convenient in operation, and more importantly it needs less amount of reagents.
