Microtomy of insect sensilla embedded in resin blocks for transmission electronic microscopy

Abstract:Transmission electronic microscopy (TEM) can be used to examine microslices of sensilla located on different insect organs thereby revealing the ultrastructure of each type of sensilla and providing evidence for their function analysis. However, it’s difficult to accurately locate different types of sensilla by mocrotomy for TEM. The precise location of insect sensilla embedded inside resin blocks was determined using both scanning electronic microscopy (HACHI S3400N) and fluorescent microscopy (Leica md2500). This new method of microtomy of insect samples in resin blocks can improve the accuracy of locating insect sensilla and save research costs.
